GV20-0251 is a fully human, Fc-attenuated IgG1 monoclonal antibody developed by GV20 Therapeutics. It targets the immune checkpoint Immunoglobulin Superfamily Member 8 (IGSF8), acting as an antagonist to inhibit its function. By binding to IGSF8 expressed on various immune cells, GV20-0251 prevents the interaction of IGSF8 with its ligands, thereby enhancing natural killer (NK) cell-mediated cytotoxicity against malignant cells, upregulating dendritic cell antigen presentation, and increasing T cell activation. The drug is being investigated primarily for advanced and/or refractory solid tumor malignancies in Phase 1/2 clinical trials as both monotherapy and in combination with pembrolizumab (Keytruda). It represents the first AI-designed antibody against an AI-predicted target to enter clinical development[2][5][6][7].
