Overview

GVAX + ipilimumab is a combination immunotherapy regimen under investigation for the treatment of various cancers, including metastatic pancreatic ductal adenocarcinoma (PDA), metastatic castration-resistant prostate cancer (mCRPC), and neuroblastoma[1][4][5][6]. GVAX refers to a family of vaccines made from irradiated allogeneic or autologous tumor cells genetically modified to secrete granulocyte-macrophage colony-stimulating factor (GM-CSF). This promotes antigen presentation and stimulates an anti-tumor immune response by recruiting and activating dendritic cells. Ipilimumab is a monoclonal antibody that targets cytotoxic T-lymphocyte-associated protein 4 (CTLA-4), blocking its inhibitory signal on T cells and thereby enhancing T-cell-mediated anti-tumor activity[3][5]. The combination aims to synergistically boost the immune system’s ability to recognize and destroy cancer cells. Clinical trials have shown biological activity in several solid tumors but mixed efficacy results; for example, in metastatic PDA maintenance therapy, the combination did not improve overall survival compared with chemotherapy continuation[5][8].
