GVAX leukemia vaccine is an autologous cell-based immunotherapy consisting of a patient's own leukemia cells that have been genetically modified to secrete Granulocyte-Macrophage Colony-Stimulating Factor (GM-CSF) and then irradiated. The vaccine is designed to stimulate a systemic immune response against leukemia-associated antigens. Upon intradermal administration, the secreted GM-CSF recruits and activates dendritic cells to the injection site. These antigen-presenting cells then ingest the irradiated tumor cells, process the full spectrum of tumor-specific antigens, and migrate to regional lymph nodes to prime T-cell responses. This approach aims to eliminate minimal residual disease and prevent relapse in patients with acute myelogenous leukemia (AML) or chronic myelogenous leukemia (CML).
