Overview

GVAX pancreatic cancer vaccine is an allogeneic whole-cell immunotherapy consisting of two irradiated pancreatic cancer cell lines (PANC 10.05 and BxPC-3) genetically modified to secrete human granulocyte-macrophage colony-stimulating factor (GM-CSF). Developed initially by Cell Genesys and later by Aduro Biotech, the vaccine is designed to stimulate a systemic T-cell-mediated immune response against pancreatic tumor antigens. GM-CSF acts as a potent recruiter and activator of dendritic cells at the injection site, which then process and present tumor-associated antigens to the immune system. It has been studied primarily in the neoadjuvant and adjuvant settings for resectable pancreatic adenocarcinoma, often in combination with low-dose cyclophosphamide to deplete regulatory T cells, or with other immunotherapies like CRS-207 or checkpoint inhibitors.
