**GVAX Prostate** is an investigational allogeneic cancer cellular vaccine for prostate cancer, consisting of irradiated prostate cancer cell lines (primarily LNCaP and PC-3) genetically modified to secrete granulocyte-macrophage colony-stimulating factor (GM-CSF). GM-CSF enhances dendritic cell recruitment and activation at the vaccination site, promoting antigen presentation from whole tumor cells to elicit both T-cell and B-cell immune responses against prostate cancer antigens, including those from lymph node and bone metastases. Developed initially by Cell Genesys, it demonstrated antitumor activity in preclinical rat and transgenic models, safety and immune responses in early clinical trials, and improved median overall survival (e.g., 26-35 months vs. predicted 19-22 months) in phase II studies of metastatic castration-resistant prostate cancer (mCRPC), though phase III trials (VITAL-1 and VITAL-2) did not meet endpoints and were halted.
