The GVGH iNTS GMMA vaccine (iNTS-GMMA) is an investigational bivalent outer membrane vesicle-based vaccine developed by the GSK Vaccines Institute for Global Health (GVGH), a division of GSK Biologicals. It is designed to prevent invasive nontyphoidal Salmonella (iNTS) disease caused by *Salmonella enterica* serovars Typhimurium and Enteritidis. The vaccine utilizes Generalized Modules for Membrane Antigens (GMMA) — outer membrane vesicles (OMVs) shed from genetically modified *Salmonella* bacteria — as a delivery platform for the O-antigen (OAg) portion of lipopolysaccharide (LPS). The parent bacteria are genetically engineered with a `tolR` deletion to induce hyperblebbing and `msbB` and `pagP` deletions to reduce lipid A acylation, thereby lowering endotoxicity and reactogenicity. The vaccine is formulated with aluminum hydroxide (Alhydrogel) and is administered intramuscularly. It is currently in Phase II clinical development, with trials evaluating its safety, reactogenicity, and immunogenicity in African populations, including infants, children, and adults.
