The GVGH Shigella sonnei 1790GAHB vaccine is an investigational prophylactic vaccine designed to prevent infection by Shigella sonnei, a major cause of bacillary dysentery (shigellosis). It is based on the Generalized Modules for Membrane Antigens (GMMA) technology, which utilizes outer membrane vesicles derived from genetically modified S. sonnei bacteria that express high levels of O antigen (OAg). The formulation contains GMMA proteins and aluminum hydroxide as an adjuvant. The primary mechanism involves stimulating the immune system to produce antibodies against the lipopolysaccharide (LPS) O antigen of S. sonnei, thereby conferring protective immunity through immunostimulation and antibody-mediated responses. Clinical studies have shown that the vaccine is immunogenic and well tolerated in adults[1][4][5][6][7]. Development has been led by GSK Vaccines Institute for Global Health (GVGH), with GlaxoSmithKline involved in manufacturing.
