Overview

GVXN SD133 is a monovalent bioconjugate vaccine candidate designed to prevent infections caused by *Shigella dysenteriae* type 1. Developed by GlycoVaxyn (now a subsidiary of GSK), the vaccine utilizes a proprietary bioconjugation platform where the *S. dysenteriae* O1 lipopolysaccharide (LPS) O-antigen is enzymatically linked to a carrier protein, typically the *Pseudomonas aeruginosa* Exoprotein A (EPA), within engineered *Escherichia coli* cells. This approach aims to overcome the limitations of traditional chemical conjugation by ensuring a more consistent and defined product. The vaccine is intended to elicit high titers of functional antibodies against the O-antigen, providing protection against bacillary dysentery. While it successfully completed Phase 1 clinical testing, development of this specific monovalent asset has been superseded by multivalent Shigella vaccine programs within the GSK pipeline.
