GW2850 is a small molecule kinase inhibitor that selectively targets the colony-stimulating factor 1 receptor (CSF1R), also known as c-Fms. Developed by GSK, it is primarily utilized in preclinical research to investigate the role of macrophage signaling within the tumor microenvironment. In models of mesothelioma, GW2850 has been shown to inhibit CSF1R signaling in tumor-associated macrophages, thereby reprogramming them from a pro-tumor (Th2) to an anti-tumor (Th1) phenotype. This modulation enhances the apoptotic response of tumor cells to standard chemotherapy agents such as cisplatin and pemetrexed. While it has demonstrated potential in enhancing chemoresponsiveness in organotypic models, it remains a tool compound used to study macrophage-mediated chemoresistance.
