Overview

GW311616A is a potent, selective, and orally active small molecule inhibitor of neutrophil elastase (NE). Originally developed by GlaxoSmithKline (GSK), it is frequently utilized as a pharmacological tool to study the role of NE in inflammatory and cardiovascular pathologies. Recent research presented at AHA 2025 highlights its potential in treating heart failure with preserved ejection fraction (HFpEF) by targeting the NE-RBPMS-Titin signaling axis. In preclinical models, GW311616A has demonstrated the ability to ameliorate diastolic dysfunction and reduce cardiac inflammation by preventing the downregulation of RNA-binding protein with multiple splicing (RBPMS), thereby preserving Titin mRNA alternative splicing and cardiac function.
