Overview

GW328267X is a small molecule drug developed as a dual adenosine receptor modulator. It acts as a potent and selective agonist at the human adenosine A2A receptor and an antagonist at the human adenosine A3 receptor. The compound was investigated for its anti-inflammatory effects in conditions such as asthma and allergic rhinitis (via inhaled or intranasal administration), but failed to show efficacy in clinical trials despite promising preclinical results. Side effects included hypotension and tachycardia related to its mechanism of action. Later studies explored intravenous administration for potential use in acute lung injury; however, dose-limiting cardiovascular side effects led to discontinuation of clinical development[1][2][3][4][6].
