Overview

GW695634 is an orally active prodrug developed by GlaxoSmithKline (now GSK) for the treatment of HIV-1 infection. It is converted in vivo to its active form, GW678248, through amide hydrolysis. The drug acts as a next-generation non-nucleoside reverse transcriptase inhibitor (NNRTI), designed to inhibit both wild-type and NNRTI-resistant strains of HIV-1—including those with common resistance mutations such as K103N and Y181C. Clinical studies demonstrated that GW695634 produced significant reductions in viral load among patients with NNRTI-resistant HIV-1 and was generally well tolerated over short-term administration. However, development was discontinued after Phase II clinical trials[2][3][5][6][7].
