GW870086 is a potent anti-inflammatory small molecule steroid developed as a selective glucocorticoid receptor agonist. It was designed to modulate only a subset of genes typically regulated by classical glucocorticoids, aiming to retain anti-inflammatory efficacy while reducing side effects. In preclinical and clinical studies, GW870086 demonstrated strong repression of inflammatory cytokine release and unique gene regulation compared to standard glucocorticoids like fluticasone propionate. It showed comparable anti-inflammatory activity in models of contact dermatitis and allergic inflammation but did not demonstrate significant clinical efficacy in asthma or atopic dermatitis trials. The drug was investigated for inhaled use in asthma and topical use in atopic dermatitis but its development has been discontinued[1][3][5][6][7][8].
