GWN323 is an investigational human IgG1 monoclonal antibody that targets the glucocorticoid-induced tumor necrosis factor receptor (GITR), a protein expressed on various T-cell subsets. By binding to and activating GITR, GWN323 stimulates the immune system, promoting activation and proliferation of tumor-antigen-specific T effector cells (Teff) while suppressing the function of activated regulatory T cells (Tregs). This dual mechanism enhances anti-tumor immune responses by shifting the balance toward increased Teff activity and reduced immunosuppression from Tregs. GWN323 is being developed primarily for advanced solid tumors and lymphomas. Preclinical studies have shown synergistic effects when combined with checkpoint inhibitors, such as PD-1 inhibitors[1][2][3][4][5].
