Overview

**GX-07-TM** is an institution-developed, myeloablative conditioning regimen used before allogeneic hematopoietic stem cell transplantation for transfusion-dependent thalassemia. The regimen combines **fludarabine**, **busulfan**, **cyclophosphamide**, and **anti-thymocyte globulin**. Busulfan and cyclophosphamide provide intensive marrow ablation through DNA damage, fludarabine adds lymphodepleting purine-analogue activity, and anti-thymocyte globulin provides broad T-cell immunosuppression to facilitate engraftment and reduce graft rejection. It was developed by the First Affiliated Hospital of Guangxi Medical University and has been evaluated in large retrospective and prospective multicenter cohorts involving matched sibling, unrelated, and haploidentical donor transplantation.
