GX-188E + GX-I7 + pembrolizumab is a triple combination therapy under investigation for the treatment of HPV type 16 or 18 positive head and neck squamous cell carcinoma (HNSCC) and other HPV-related cancers. GX-188E is a therapeutic DNA vaccine encoding the E6/E7 fusion protein of human papillomavirus (HPV) subtypes 16 and 18, designed to induce immune responses against tumor cells expressing these viral proteins. It is administered intramuscularly by electroporation[7]. GX-I7 (efineptakin alfa, rhIL-7-hyFc) is a long‐acting recombinant human interleukin‐7 fused to an Fc domain, developed to correct T-cell deficiency and amplify anti-tumor immune responses by increasing CD4+ and CD8+ T cell numbers[6][8]. Pembrolizumab (Keytruda) is a monoclonal antibody targeting programmed death receptor 1 (PD‑1), functioning as an immune checkpoint inhibitor to enhance anti-tumor immunity. The combination aims to synergistically boost anti-tumor immunity through vaccination-induced antigen presentation, T-cell amplification, and checkpoint inhibition. Early-phase clinical trials have shown promising activity with manageable safety in patients with resectable HPV-positive HNSCC[1][2][3].
