Overview

GX1 is a synthetic seven-amino acid cyclic peptide (sequence: Tyr-Glu-Gln-Asp-Pro-Trp-Gly) that serves as a high-affinity targeting ligand for the neovasculature of gastric cancer. Identified through in vivo phage display technology, GX1 specifically recognizes and binds to transgelin (TAGLN), a protein significantly upregulated in the vascular endothelial cells of gastric tumors compared to normal gastric tissue. By acting as a homing agent, GX1 is utilized to deliver various therapeutic payloads—including radionuclides, chemotherapeutic nanoparticles, and pro-apoptotic proteins like mutant human tumor necrosis factor (rmhTNF)—directly to the tumor site. This targeted approach aims to enhance the local concentration of anti-cancer agents while minimizing systemic side effects, particularly in the treatment of advanced gastric carcinoma.
