Overview

GZ‑389988 is a small molecule investigational drug developed for the treatment of osteoarthritis pain. It acts as a selective antagonist of the tropomyosin receptor kinase A (TrkA), which is the high-affinity receptor for nerve growth factor (NGF). By inhibiting TrkA, GZ‑389988 blocks NGF/Trk pathway activation, thereby reducing pain signaling associated with osteoarthritis. The compound was originally developed by Genzyme and later by Sanofi following acquisition. Clinical development included phase II trials using intra-articular injection in patients with knee osteoarthritis, where it demonstrated some efficacy in reducing pain but was associated with injection site inflammatory reactions. Development for osteoarthritis has since been discontinued[1][2][4][5][6].
