GZ21T

**GZ21T** is an investigational topical cream developed by Genzada Pharmaceuticals for dermatologic use, principally actinic keratosis. It is a fixed-ratio topical formulation of the three small-molecule active ingredients **isovanillin, harmine, and curcumin**, corresponding to the active-ingredient combination used in the oral product GZ17-6.02. Preclinical actinic-keratosis studies found that GZ21T reduced lesion burden and suppressed signaling associated with cellular survival and proliferation, including MAPK, PI3K-AKT, HIF-1alpha, Wnt, insulin, and ERBB pathways. Cellular studies also support induction of endoplasmic-reticulum stress, AMPK and ULK1 activation, autophagy, and death-receptor-associated cell death, with suppression of AKT, mTORC1, and YAP signaling. A placebo-controlled Phase 1 study evaluated single ascending topical doses in healthy volunteers.
