Overview

H-ferritin siRNA is a small interfering RNA therapeutic candidate designed to silence the expression of the ferritin heavy chain (FTH1) gene. Ferritin is primarily known as an intracellular iron storage protein, but it also localizes to the nuclei of tumor cells, where it protects DNA from oxidative damage and facilitates transcription. By knocking down H-ferritin levels, this siRNA disrupts iron homeostasis and increases the sensitivity of cancer cells, such as glioma, to DNA-alkylating chemotherapeutic agents like carmustine (BCNU). Preclinical studies have demonstrated that delivery via cationic liposomes can significantly reduce the effective dose of chemotherapy required for tumor suppression and increase apoptosis in vitro and in vivo.
