Overview

H-His-D-Arg-Ala-Trp-D-Phe-Lys-NH2 is a synthetic hexapeptide that acts as a competitive antagonist of alpha-melanocyte-stimulating hormone (alpha-MSH). It was developed as a pharmacological tool to study the melanocortin system, specifically targeting the melanocortin 1 receptor (MC1R). The peptide is structurally related to growth hormone-releasing peptides (GHRPs) but contains a D-Arginine substitution at the second position, which eliminates growth hormone-releasing activity and confers potent MSH-antagonistic properties. Research has demonstrated its ability to inhibit MSH-induced skin darkening in various vertebrate species, such as the leopard frog (*Rana pipiens*) and the tree lizard (*Urosaurus ornatus*). However, its biological activity is highly species-specific, as it has been found to be inactive in certain mammalian systems, including murine melanoma cells.
