ISIS 2503 is a synthetic, 20-base phosphorothioate antisense oligodeoxynucleotide designed to hybridize to the translation initiation region of human H-Ras mRNA. By binding specifically to this sequence, it selectively inhibits the expression of H-Ras, an oncogene implicated in the development and maintenance of various cancers. This targeted inhibition may suppress tumor cell growth in Ras-dependent tumors while sparing other members of the Ras protein family. Developed as an anti-cancer agent, ISIS 2503 has been evaluated primarily for pancreatic cancer and also investigated in metastatic breast cancer and non-small cell lung cancer. The drug was developed by Isis Pharmaceuticals (now Ionis Pharmaceuticals) and reached Phase II clinical trials[1][2][3][5][7].
