Overview

The H1299 cell lysate vaccine is an investigational cancer vaccine developed by the National Cancer Institute (NCI) for the treatment of locally advanced esophageal cancer. The vaccine is composed of lysates derived from the H1299 non-small cell lung carcinoma cell line, which is known to express high levels of cancer-testis (CT) antigens, including NY-ESO-1 and various CT-X genes. These antigens are typically restricted to germ cells but are aberrantly expressed in various malignancies, making them ideal targets for immunotherapy. The vaccine is formulated with the Montanide ISA-51 VG adjuvant and administered via subcutaneous injection. It is currently being evaluated in Phase I/II clinical trials (NCT05898828) as an adjuvant therapy for patients with Stage II or III esophageal cancer who have residual disease after neoadjuvant chemoradiation. In these trials, it is often combined with the histone deacetylase (HDAC) inhibitor entinostat and the PD-1 checkpoint inhibitor nivolumab to enhance the immune response and counteract tumor-mediated immunosuppression.
