H1299 lysate vaccine is an investigational cancer vaccine biologic developed by the National Cancer Institute (NCI). It is composed of lysates derived from the H1299 large cell lung cancer cell line, which is characterized by high expression of various cancer-testis antigens (CTAs), including NY-ESO-1, MAGE-A1, and MAGE-A3. The vaccine is designed to stimulate the host immune system to recognize and attack tumor cells expressing these antigens, which are typically restricted to germ cells in the testes and placenta but aberrantly expressed in various malignancies. It is typically administered via deep intradermal or subcutaneous injection alongside adjuvants such as Iscomatrix or Montanide ISA-51 VG to enhance the immunogenic response. Clinical evaluation has primarily focused on thoracic malignancies, including non-small cell lung cancer and esophageal cancer.
