h15B6 CAR-T is a humanized chimeric antigen receptor T cell therapy targeting the juxtamembrane region of mesothelin (MSLN), developed to overcome tumor resistance caused by shed mesothelin. The therapy utilizes an antibody (15B6) engineered to bind specifically to a membrane-proximal, protease-sensitive region of mesothelin that is retained on cancer cells but not present on shed forms of the protein, preventing off-target binding and thus enhancing specificity and efficacy. In preclinical models, h15B6 CAR-T demonstrated complete and durable tumor regressions—especially in hard-to-treat pancreatic cancer xenografts—by effectively killing mesothelin-expressing tumor cells and expanding in the tumor environment. This addresses a key limitation of prior mesothelin-targeted CAR-T constructs, which were inhibited by shed mesothelin binding[1][3][5].
