Overview

h15H3-vcMMAE is a preclinical antibody-drug conjugate (ADC) developed by Seattle Genetics (now Pfizer) that targets integrin β6 (ITGB6). The agent consists of a humanized monoclonal antibody, h15H3, conjugated to the microtubule-disrupting cytotoxin monomethyl auristatin E (MMAE) via a protease-cleavable valine-citrulline (vc) linker. Integrin β6 is an epithelial-specific integrin that is significantly upregulated in various solid tumors, including those of the pancreas, head and neck, lung, and bladder, while maintaining low expression in healthy tissues. Upon binding to the cell surface, the ADC is internalized and trafficked to lysosomes, where the linker is cleaved to release MMAE, leading to cell cycle arrest and apoptosis.
