Overview

H19 siRNA is a small interfering RNA (siRNA) designed to silence the expression of the long non-coding RNA (lncRNA) H19. H19 is a developmentally regulated, imprinted gene that is frequently overexpressed in various malignancies and inflammatory conditions. By targeting H19 for degradation through the RNA-induced silencing complex (RISC), H19 siRNA modulates multiple downstream signaling pathways, including AKT, NF-κB, JNK, and the p53/Bax/Bcl-2 axis. It also acts by disrupting the "sponging" effect of H19 on various microRNAs, such as miR-675, miR-29a, and miR-199a. Preclinical studies have demonstrated that H19 siRNA can inhibit tumor cell proliferation, migration, and invasion, reduce neuroinflammation in stroke models, alleviate neuropathic pain, and improve insulin sensitivity in diabetic models. Currently, H19 siRNA is primarily utilized as a research tool in preclinical investigations across oncology, cardiology, and neurology.
