H1N1 SynCon is an investigational synthetic consensus DNA vaccine directed against the hemagglutinin antigen of influenza A H1N1, designed using Inovio’s SynCon technology to provide broad, “universal” cross-strain protection within the H1 subtype. The vaccine encodes a synthetic consensus H1 antigen derived from multiple existing H1N1 strains, with the aim of eliciting humoral and cellular immune responses that protect against both historical and newly emergent H1N1 viruses, rather than matching a single circulating strain.[1][2] In preclinical studies, the H1N1 SynCon vaccine provided 100% protection in animals challenged with both the 2009 swine-origin H1N1 virus and the 1918 pandemic H1N1 virus and generated protective hemagglutination inhibition titers against additional H1N1 strains.[1][2] The construct and related DNA vaccines have been developed by Inovio in collaboration with the University of Pennsylvania and are delivered using Inovio’s electroporation technology as part of broader universal influenza vaccine programs.[1][2]
