H1ssF (VRC-FLUNPF099-00-VP) is an investigational influenza vaccine designed to induce broadly neutralizing antibodies by targeting the conserved stem region of the hemagglutinin (HA) protein from the influenza A/H1N1 virus. The vaccine consists of a stabilized HA stem domain from A/New Caledonia/20/1999 fused to a ferritin nanoparticle scaffold, which presents multiple copies of the HA stem in a repetitive array. This design aims to focus immune responses on the less variable HA stem rather than the immunodominant head, potentially providing cross-protection against diverse group 1 influenza strains. Preclinical studies demonstrated that H1ssF elicits robust and broad antibody responses and confers protection against heterosubtypic challenge in animal models. The primary indication is for prevention of influenza A infection; it is being developed as a universal or broadly protective flu vaccine[1][2][3].
