Overview

h2CAR-T is a novel humanized chimeric antigen receptor (CAR) T-cell therapy targeting B-cell maturation antigen (BCMA), developed for the treatment of relapsed or refractory multiple myeloma. Developed through a collaboration between the Indian Institute of Technology Bombay, Tata Memorial Centre, and ImmunoACT, h2CAR-T utilizes a humanized scFv to reduce immunogenicity and inflammatory toxicities such as cytokine release syndrome (CRS). In preclinical studies, h2CAR-T demonstrated high binding affinity (KD: 0.55nM), robust in vitro cytotoxicity against BCMA-expressing cells, and superior in vivo tumor suppression compared to murine-based CAR-T cells. Notably, it exhibited lower secretion of pro-inflammatory cytokines (IFN-γ, TNF-α, IL-2, IL-6, and IL-1β) and a stable memory phenotype with reduced exhaustion markers, supporting its progression into first-in-human clinical trials.
