H5-VLP + GLA-AF is an investigational combination vaccine developed for the prevention of pandemic influenza caused by the H5N1 subtype. The formulation consists of a recombinant hemagglutinin (H5) protein derived from the A/Indonesia/5/2005 strain, produced in plants using a transient expression system to assemble into virus-like particles (VLPs). This antigen is combined with GLA-AF, an adjuvant based on glucopyranosyl lipid A formulated in aqueous form. The VLP component mimics native viral structure to elicit strong immune responses without containing infectious material, while GLA-AF acts as a potent immunostimulant targeting Toll-like receptor 4 (TLR4), enhancing both humoral and cellular immunity. Clinical studies have shown that this combination increases immunogenicity and broadens protection against divergent H5N1 strains when administered intradermally or intramuscularly[1][2][6]. Developed primarily by Medicago in collaboration with IDRI, it has reached Phase 2 clinical trials for human influenza prevention[4][6].
