**H5.010RSVTK** is an investigational, replication-defective adenoviral gene-therapy vector that delivers the **herpes simplex virus thymidine kinase** transgene under control of the Rous sarcoma virus promoter. It was administered by stereotactic intratumoral injection for malignant brain tumors, followed by separately administered ganciclovir. Transduced tumor cells express HSV-TK, which converts ganciclovir into cytotoxic phosphorylated metabolites, creating a prodrug-activation or suicide-gene-therapy strategy intended to kill proliferating tumor cells.
