h5G1.1-mAb is an experimental humanized monoclonal antibody designed to target the C5 component of the complement system. Developed by Alexion Pharmaceuticals in collaboration with the National Institute of Arthritis and Musculoskeletal and Skin Diseases (NIAMS), it was specifically investigated for the treatment of dermatomyositis. The antibody works by binding to C5 and blocking its cleavage into the pro-inflammatory mediator C5a and the membrane attack complex (C5b-9), which is implicated in the capillary damage and muscle inflammation characteristic of dermatomyositis. h5G1.1-mAb was created by humanizing a murine anti-C5 antibody (m5G1.1-mAb) and served as the precursor molecule to eculizumab (Soliris), a long-acting version of the same antibody. Its clinical evaluation included a Phase 2 randomized, double-blind, placebo-controlled pilot study in adult patients with persistent dermatomyositis.
