A self-assembling ferritin-based nanoparticle influenza vaccine that displays multiple copies of influenza A H5 hemagglutinin (HA) on a ferritin core to enhance B cell activation and elicit broad neutralizing antibody responses, particularly to conserved HA epitopes. Ferritin nanoparticles present ordered, multivalent HA, driving stronger germinal center reactions and improved immunogenicity versus soluble HA. Related ferritin HA vaccines from the same platform have shown safety and immunogenicity in phase 1 clinical trials (e.g., full-length H2HA-ferritin and HA stem constructs), with heterosubtypic protection against H5N1 in animals when using a group 1 HA stem on ferritin. Primary indication is prevention of influenza A (H5N1) infection and pandemic preparedness. Key developers of the ferritin HA nanoparticle platform include the Vaccine Research Center at NIAID/NIH, with manufacturing commonly via recombinant expression systems such as insect cell–baculovirus for HA-ferritin assembly.[6][1][8]
