Drug intelligence / Profile preview
H5N1 vaccine AF03 adjuvanted is an inactivated split-virion influenza A(H5N1) vaccine formulated with Sanofi’s AF03 oil-in-water emulsion adjuvant to enhance and broaden immune responses against avian H5N1 strains while enabling antigen dose sparing. The AF03 adjuvant contains squalene with surfactants such as sorbitan oleate and polyoxyethylene cetostearyl ether in an aqueous phase with mannitol, and is mixed with low-dose hemagglutinin antigen from H5N1 strains such as A/Indonesia/5/2005 in a two-vial presentation before intramuscular injection.[7][8] AF03 enhances antigen presentation and promotes strong humoral responses, increasing seroconversion and seroprotection rates at reduced HA doses compared with non-adjuvanted formulations, and has been evaluated in adult phase 2 clinical trials for pandemic preparedness.[2][8]
