HA + DOX-COF is a nanomedicine drug delivery system composed of doxorubicin (DOX) loaded into a covalent organic framework (COF), with the surface modified by hyaluronic acid (HA). This formulation is designed to enhance targeted delivery of doxorubicin to tumor cells. The COF provides high porosity and stability for efficient drug loading and controlled release. Hyaluronic acid modification enables active targeting via binding to CD44 receptors, which are overexpressed on many cancer cell surfaces, thereby increasing tumor-specific accumulation. The system exhibits pH and glutathione-responsive drug release properties, ensuring that doxorubicin is preferentially released in the tumor microenvironment while reducing toxicity to healthy tissues. The primary mechanism of action derives from doxorubicin’s DNA intercalation and inhibition of topoisomerase II, leading to DNA damage and apoptosis in cancer cells[1][2][3].
