HA-22-31 is a small molecule inhibitor of chloride intracellular channel 3 (CLIC3) currently in preclinical development for the treatment of metastatic breast cancer. CLIC3 is secreted by cancer cells and cancer-associated fibroblasts, promoting tumor invasion and angiogenesis through its oxidoreductase activity, which inhibits transglutaminase-2 (TGM2). HA-22-31 was identified via an in silico screen of over 10 million small molecules and subsequent structure-activity relationship (SAR) analysis. It directly binds to CLIC3 with an IC50 of 1.7 μM, significantly inhibiting cell migration and tumor metastasis in preclinical models.
