Overview

HA-irinotecan is a targeted formulation of the cytotoxic agent irinotecan, developed using Alchemia's HyACT (Hyaluronic Acid Chemotherapy Targeting) platform. The drug consists of irinotecan non-covalently formulated with hyaluronic acid (HA), a naturally occurring polymer that binds to the CD44 receptor. Since CD44 is frequently overexpressed in various solid tumors, including colorectal cancer, the HA component is designed to selectively deliver irinotecan to the tumor site, potentially enhancing efficacy and reducing systemic side effects. In a pivotal Phase 3 trial (COL-003) for metastatic colorectal cancer, HA-irinotecan failed to demonstrate a statistically significant improvement in progression-free survival compared to standard irinotecan, leading to the discontinuation of its primary development program by Alchemia and its subsequent acquisition by Panther Biotechnology.
