Overview

The HA2 peptide vaccine is an investigational peptide-based therapeutic vaccine developed by Hannover Medical School (Medizinische Hochschule Hannover) in Germany. Its active ingredient is the immunogenic HA-2 (minor histocompatibility antigen HA-2, specifically the HA-2V variant) peptide, which is an HLA-A2-restricted minor histocompatibility antigen (mHAg) derived from a biallelic locus. The vaccine has no known commercial brand name, as it is a research-stage compound. Its biological target is the minor histocompatibility antigen HA-2, which is expressed on hematopoietic cells and triggers HLA-A2-restricted alloreactive cytotoxic T-cell responses. The modality is a peptide vaccine (biologic). It was evaluated in a Phase I/II clinical trial in Germany in adults with advanced high-risk leukemia or leukemic relapse who had undergone allogeneic hematopoietic stem cell transplantation (HSCT) and had donor-recipient HA-2 disparity. The primary objective of the trial was to assess immunization toxicity, with relapse prevention as a secondary objective. The vaccine was administered post-transplant to harness the graft-versus-leukemia (GvL) effect by stimulating donor-derived T cells to selectively target residual malignant hematopoietic cells expressing the HA-2 antigen. The study also included vaccination with the related HA-1 peptide in eligible patients.
