Humanized AB21 (hAB21) is a pan-allelic anti-SIRPα antibody that blocks the interaction between Signal regulatory protein α (SIRPα) and its ligand CD47. This blockade enhances macrophage-mediated antibody-dependent phagocytosis of tumor cells and promotes anti-tumor activity. It has demonstrated improved responses in combination with rituximab in mouse xenograft models and with PD-1/PD-L1 blockade in syngeneic models, leading to the activation of monocytes, dendritic cells, and T cell effector functions. hAB21 is being developed as a potential therapeutic intervention for human malignancies.
