Overview

**HACE-MbCD** is an experimental CD44-targeted, cholesterol-depleting polymeric nanoassembly developed for **breast cancer** therapy. It consists of methyl-β-cyclodextrin covalently conjugated through an ester linkage to a hyaluronic acid-ceramide amphiphile, which self-assembles into approximately 198 nm nanoparticles. The hyaluronic acid component promotes tumor-cell association through CD44, while the methyl-β-cyclodextrin moiety extracts membrane cholesterol and disrupts cholesterol-rich lipid rafts. In MDA-MB-231 breast cancer cells and tumor-bearing mice, intravenous HACE-MbCD nanoassembly increased lipid-raft disruption, apoptosis, tumor targeting, and tumor-growth suppression relative to unconjugated methyl-β-cyclodextrin or HACE nanoassembly controls. It remains a preclinical research-stage therapeutic system.
