hAd5-HPV vaccine (IBRX-042) is a second-generation adenovirus-based therapeutic vaccine developed by ImmunityBio. It utilizes a human adenovirus serotype 5 (hAd5) vector that has been modified with deletions in the E1, E2b, and E3 regions. These modifications are intended to reduce the vector's immunogenicity, thereby minimizing the host's anti-vector immune response and allowing for more effective repeated dosing. The vaccine is engineered to express the E6 and E7 oncoproteins of Human Papillomavirus (HPV) types 16 and 18. By delivering these antigens, the vaccine aims to stimulate a potent T-cell mediated immune response against HPV-infected and malignant cells. It is currently being evaluated in clinical trials for HPV-associated malignancies, including oropharyngeal squamous cell carcinoma and cervical cancer, often as part of a combination regimen with the IL-15 superagonist nogapendekin alfa inbakicept (NAI).
