hAd5-S-Fusion + N-ETSD is a dual-antigen COVID-19 vaccine candidate delivered by a next-generation human adenovirus serotype 5 (hAd5) vector with E1, E2b, and E3 deletions. It encodes the SARS-CoV-2 spike (S) protein, modified for enhanced cell surface display (S-Fusion), to increase humoral responses, and the viral nucleocapsid (N) protein fused with an Enhanced T-cell Stimulation Domain (N-ETSD) to direct N to the endosomal/lysosomal compartment for enhanced MHC I and II antigen presentation. This formulation is designed to generate strong and broad immune responses, including neutralizing antibodies and robust CD4+ and CD8+ T-cell activation toward both S and N antigens, with a Th1-bias. The vaccine is available in subcutaneous, intranasal, and oral formulations, with the oral form noted for thermal stability and avoiding the need for ultra-cold storage. Developed primarily for COVID-19, it is in clinical trials and preclinical models, showing protection in both upper and lower respiratory tracts in animal studies and in vitro activation of human T cells[1][2][3].
