The ImmunityBio COVID-19 oral vaccine (hAd5-S-N) is a second-generation, non-replicating human adenovirus serotype 5 (hAd5) vector-based vaccine developed by ImmunityBio. It is a bivalent construct that expresses both the SARS-CoV-2 spike (S) and nucleocapsid (N) proteins, designed to elicit both robust T-cell and B-cell immune responses. The oral formulation utilizes iosBio's OraPro thermal stabilization technology, which involves enteric-coated capsules that protect the vaccine from stomach acid and deliver it to the small intestine. This route targets gut-associated lymphoid tissue (GALT) to induce mucosal IgA responses in addition to systemic immunity. The vaccine is stable at room temperature and is being evaluated as both a primary series and a universal booster to other spike-based vaccines.
