hAd5 vaccine is an investigational **viral vector COVID-19 vaccine** developed by ImmunityBio that uses a second-generation human adenovirus serotype 5 vector with deletions in **E1, E2b, and E3** to reduce the impact of pre-existing adenovirus immunity. The construct encodes two SARS-CoV-2 antigens: a modified **spike protein** designed to enhance cell-surface expression (**S-Fusion**) and a **nucleocapsid protein** engineered with an Enhanced T-cell Stimulation Domain (**N-ETSD**) to promote antigen presentation and T-cell priming. The program was evaluated in subcutaneous and oral formulations in Phase 1 and Phase 2 studies for prevention of COVID-19, with the goal of inducing both neutralizing antibody responses and broad cellular immunity against SARS-CoV-2.
