Haduvio is an investigational, oral extended-release formulation of nalbuphine developed for the treatment of chronic cough in patients with idiopathic pulmonary fibrosis (IPF), refractory chronic cough (RCC), and pruritus associated with prurigo nodularis. Its mechanism of action is unique among antitussive therapies, acting as a kappa opioid receptor agonist and mu opioid receptor antagonist (KAMA). This dual action targets both central and peripheral components of the cough reflex arc, aiming to reduce cough hypersensitivity regardless of the initial trigger. Haduvio has demonstrated statistically significant reductions in 24-hour cough frequency in clinical trials for RCC and IPF-related chronic cough. The drug is being developed by Trevi Therapeutics and has not yet been approved for any indication[1][3][4][8].
