Overview

This is a combination regimen consisting of hepatic arterial infusion chemotherapy (HAIC), a programmed cell death protein 1 (PD-1) immune checkpoint inhibitor, and the multikinase inhibitor lenvatinib. - **HAIC** delivers chemotherapeutic agents directly into the liver via the hepatic artery to maximize local drug concentration for hepatocellular carcinoma (HCC). - **PD-1 inhibitors** are monoclonal antibodies that block the interaction between PD-1 on T cells and its ligands (PD-L1/PD-L2), thereby enhancing antitumor immune responses by preventing tumor-induced T-cell inhibition. - **Lenvatinib** is an oral small molecule tyrosine kinase inhibitor targeting multiple receptors including VEGFRs, FGFRs, RET, KIT, and PDGFRα; it inhibits angiogenesis and tumor proliferation. The triple combination has shown improved objective response rates (ORR), progression-free survival (PFS), overall survival (OS), and disease control rate in advanced or unresectable hepatocellular carcinoma compared to monotherapies or dual combinations[2][4]. The regimen is under investigation in clinical trials for advanced HCC[4].
