Overview

This is a combination therapy consisting of hepatic arterial infusion chemotherapy (HAIC), the STRIDE regimen, and lenalidomide. - **HAIC** is a locoregional chemotherapy technique where anticancer drugs are delivered directly into the hepatic artery to treat liver tumors, most commonly hepatocellular carcinoma. The specific chemotherapeutic agents used in HAIC can vary but often include drugs like cisplatin or 5-fluorouracil. - **STRIDE** typically refers to a regimen combining durvalumab (an anti–PD-L1 monoclonal antibody) with tremelimumab (an anti–CTLA-4 monoclonal antibody), used as immunotherapy for advanced liver cancer. - **Lenalidomide** is an oral immunomodulatory drug with antineoplastic, antiangiogenic, and immune-stimulating properties. It acts by modulating cereblon E3 ubiquitin ligase complex activity, leading to tumor cell apoptosis and enhanced immune response[5][6][8][10]. This combination aims to leverage local cytotoxic effects on liver tumors (via HAIC), systemic immune activation against cancer cells (via STRIDE), and additional antitumor/immune modulation from lenalidomide.
