Drug intelligence / Profile preview
HaloCAR T cells are a universal, programmable chimeric antigen receptor (CAR) T-cell therapy platform developed by researchers at the University of Pittsburgh. Unlike traditional CAR-T therapies that utilize a single-chain variable fragment (scFv) to target a specific antigen, HaloCAR T cells express a CAR containing HaloTag, a self-labeling haloalkane dehalogenase enzyme. This system is programmed and armed in vivo or in vitro using tumor-targeting adaptors (such as antibodies or small molecules) conjugated to a chloroalkane (CA) motif. Upon co-administration, the chloroalkane-conjugated adaptors covalently bind to the HaloTag on the CAR-T cells, enabling specific and tunable targeting of multiple tumor antigens, including CD20, HER2, EGFR, CAIX, and folate receptors, to overcome antigen heterogeneity in solid and hematological tumors.
