Overview

HAMNO (2-hydroxyamino-1,4-naphthoquinone) is a small molecule inhibitor of Replication Protein A (RPA), a critical single-stranded DNA (ssDNA)-binding protein complex involved in DNA replication, repair, and recombination. Specifically, HAMNO acts as a protein-protein interaction (PPI) inhibitor targeting the RPA1 subunit. By disrupting RPA function, the drug interferes with the DNA damage response (DDR), leading to increased sensitivity to DNA-damaging agents such as platinum-based chemotherapy and PARP inhibitors. Preclinical studies in epithelial ovarian cancer (EOC) have demonstrated that HAMNO can overcome platinum resistance and exhibits monotherapy cytotoxicity in both sensitive and resistant cell lines, suggesting its potential as a precision oncology strategy in high-grade serous ovarian cancers.
